Epitopes and Mimotopes Identification Using Phage Display for Vaccine Development against Infectious Pathogens

چکیده

Traditional vaccines use inactivated or weakened forms of pathogens which could have side effects and inadequate immune responses. To overcome these challenges, phage display has emerged as a valuable tool for identifying specific epitopes that be used in vaccines. This review emphasizes the direct connection between epitope identification vaccine development, filling crucial gap field. technique allows to engineered effectively stimulate system by presenting carefully selected epitopes. Phage involves screening libraries random peptides gene/genome fragments using serum samples from infected, convalescent, vaccinated individuals. method been identify various including SARS-CoV-2, Mycobacterium tuberculosis, hepatitis viruses, H5N1, HIV-1, Human T-lymphotropic virus 1, Plasmodium falciparum, Trypanosoma cruzi, Dirofilaria repens. Bacteriophages offer advantages such being immunogenic carriers, low production costs, customization options, making them promising alternative traditional The purpose this study highlight an approach encompasses entire process single technique, without requiring additional manipulation. Unlike conventional methods, demonstrates exceptional efficiency speed, provide significant critical scenarios pandemics.

Immunization of Sheep with Phage Mimotopes against Dermatophilosis

Random peptide libraries (RPL) displayed on the surface of filamentous bacteriophages have been extensively used as a tool to map epitopes or to identify antigenic mimics (mimotpoes) of disease-specific monoclonal antibodies or polyclonal sera. These RPL are engineered by the insertion of degenerate oligonucleotides, encoding a specific number of random amino acids, in frame with a bacteriophag...

Automated Detection of Conformational Epitopes Using Phage Display Peptide Sequences

BACKGROUND Precise determination of conformational epitopes of neutralizing antibodies represents a key step in the rational design of novel vaccines. A powerful experimental method to gain insights on the physical chemical nature of conformational epitopes is the selection of linear peptides that bind with high affinities to a monoclonal antibody of interest by phage display technology. Howeve...

Characterization of Novel Fragment Antibodies Against TNF-alpha Isolated Using Phage Display Technique

Tumor necrosis factor alpha (TNF-α) is an inflammatory cytokine which plays crucial roles in pathogenesis of inflammatory diseases. The current study aimed to investigate the binding abilities of I44 and I49 domain antibodies to TNF-α. The dAbs were expressed in bacterial expression system and purified by affinity chromatography using Ni-sepharose column. The expression and purity of the protei...
